Single and multiple early afterdepolarization caused by nickel in rat atrial muscle.

چکیده
We examined the possible role of the Na-Ca exchange (NCX) in the arrhythmogenesis in rat atrial preparations applying microelectrode technique. In control Tyrode solution preparations isolated from the sinoatrial area contracted with frequency of 48+/-4 min(-1) (group I) or 84+/-7 min(-1) (group II). In preparation beating with low frequency partial inhibition of NCX by administration of Ni2+ (0.3 mmol/l) to the bath solution caused single early afterdepolarization (EAD) on the 15th min. During the following five minutes they were transformed into multiple EADs from 4 to 47 (action potentials) with general duration of 1-12 s. The effects were reversible. Ni2+ (0.3 mmol/l) in the preparations beating with higher rate (group II) did not cause multiple EADs, but after higher Ni2+ concentration (0.5 mmol/l) single EAD was observed more often. It was concluded that Ca2+ overload due to partial block of the NCX can contribute to the development of atrial tachyarrhythmias.

Background: Noscapine is a phtalideisoquinoline alkaloid with known anti-tussive properties. Recently, some other pharmacological effects and new potential clinical applications have been proposed for the drug. Since there is no data regarding its cardiovascular effects, cardiovascular actions of Noscapine were investigated in the present study. Methods: In this experimental study, the effe...
متن کاملBRIEF COMMUNICATIONS The Time Course of Action Potential Repolarization Affects Delayed Afterdepolarization Amplitude in Atrial Fibers of the Canine Coronary Sinus
The effects of changing the time course of action potential repolarization on the amplitude and coupling interval of delayed afterdepolarizations were studied in small preparations of coronary sinus atrial fibers exposed to catecholamines. Repolarization was accelerated or retarded by current pulses passed through an intracellular microelectrode in the depolarizing or repolarizing direction. Ac...

متن کاملENDOGENOUS RELEASE OF OPIATES BY REPETITIVE ELECTRICAL FIELD STIMULATION IN THE GUINEA-PIG AND RAT ILEAL LONGITUDINAL MUSCLE
The effect of repetitive electrical field stimulation and the response of the guinea-pig and rat ileal longitudinal muscle to single pulse stimulations was examined. Single pulse field stimulation produced twitch contraction which was inhibited by repetitive field stimulation (10 Hz, 40V, 0.5 msec for 5 m). This inhibition was largely, though never completely, reversed by naloxone. Contrac...
متن کاملComparison of Hyperpolarization-Activiated Current “if ” on the Rate of Spontaneous Activity and Cycle Length before and after Cutting of Atrial Muscle away from Intact Sinoatrial Node of Rabbit
It has been shown that the hyperpolarization-activated current “if” that is blocked by 2 mM Cs+ plays a minor role on pacemaker activity of the center and a major role on activity of the periphery of rabbit intact sino-atrial node. On the other hand some investigations showed that if the atrial muscle, surrounding the sino-atrial node, is cut away there is a shift in leading pacemaker site from...
